Does the Wine Count?
Yes. The usual practice is to tip on the whole pre-tax bill, drinks included. A $100 check that includes a $40 bottle still gets a tip of $18 to $20. The server opened, poured and kept track of the bottle along with everything else.
Opinion splits only on very expensive bottles, where some diners tip a lower percentage on the wine because the work does not grow with the price. That is a judgement call rather than a rule. If a sommelier helped you choose, it is customary to thank them separately, often with cash.
